Most effective way to score points in 5* arena? I keep feeling like I could be scoring more points with my 5*. I have 66, most at r2, one at r5, 7 at r4, 10 at r3. I get about 1.8 mil points each run

Make sure your arena teams maximise the number of synergies you get for each team of 3 champions. Synergy bonuses give you more points per fight.

Masteries also affect points, albeit to a much smaller degree. The typical set up is the suicides mastery, though I think the real benefit in that is the greater speed in clearing the fights; the point increase is rather minimal in my opinion.
