I saved my masteries no suicides. But if I make match suicides is on?

    You need to apply the changed masteries through the settings "Champions deck > Masteries."

    Normally yes, but it is bugged right now and under investigation. Saved masteries don’t seem to be applying and it’s using current masteries.
