Warlock was probably the second or third champ I used the most when I went for TB, he just has so much utility, and pretty decent damage (nothing too crazy of course you won't see huge numbers but it's decent enough).
Spot(have generic awakening with about 30sig stones)
A lot of people voted for spot, but honestly having him as a 6* R3, he’s not YouTuber thumbnail face levels of great, he has great immunities, and can completely slam healing within 10-15 seconds, but other than that, he is really just a bland, margarine character with mid damage output
Keep him at R2 for the time being, and don’t make the fool’s mistake of only investing in 3-4 characters, personally, I would take up spot and venom to R2 and farm resources for 1-2 months, being at TB level I can afford about 1-2 R3’s a month, so just keep patient for now

As someone with a rank 3 6* Warlock, Rank 3 6* Sig 120 Spot and even a 5* rank 5 Warlock, I have to disagree. Warlock's control over fights with little to no effort required is just too powerful to be overlooked. Spot is great, but aside from BGs or AW defense, he really won't shine much in content.
I do agree that going for more rank 2's is always the best strategy, but getting Thronebreaker will make getting rankup materials so much easier, and Warlock kicks ass at a lot of Act 7 and TB EQ paths. He's the obvious choice. Just take Warlock to rank 3 then focus on diversifying with rank 2's.
Out of the ones listed, I think Warlock is the best play. He has served me well for so long. He was one of my absolute first r3s back in the day. Second or third.
However, I also want to mention both Venom and Venompool. Both are great champions. Venom is super useful against all kinds of #spiderverse heroes (as well as a lot of other champions in general as long as they bleed), while Venompool is great at fighting #robots. I use VP all the time in BG despite only having him at r3 and he usually comes out with 100% health since he heals like a madman as long as there are armor ups to snack on.
SpiderHam will come handy later too. Very less champs are increasingly immune to degen. Plus, the damage with porker poppers is incredible. For tech, go for either warlock or Ultron. For mutant Omega Red or Og storm.

Toad
Venom
Red Hulk
