Definitely explore acts 4 and 5. It won't quite be so good in terms of rewards, but it's a decent chunk of units which is always nice. Plus exploring act 4 gives you a free mastery point if I remember right.
What you achieve before pushing for thronebreaker is entirely up to you. I will say that pushing through the rest of act 6 is harder than most of the rest of the content in the game, including both act 7 and act 8.1. I used the cavalier monthly quest as a metric. If I could complete it easily (minimal revives) perhaps it was time to start pushing act 6 toward Grandmaster. If it still took me a boatload of revives every month, I instead focused my attention on that, getting the shards and catalysts and working to improve my roster. More 6*s aren't necessary for act 6, but they make it a hell of a lot easier.
I can give specific champion recommendations too if you want. I know there are several tough fights in 6.2 and the rest.
6.2.2 Mr. Sinister: Caustic temper, he applies poison to you and you need Fury to deal any damage. Congratulations, you have an excellent counter for him already. Captain Marvel can carry through that fight.
6.2.6 Champion: Annoying little @$^+! who gains Unstoppable a bunch and tests the limits of your sanity in that last 10%. Thing may help during that last 10% if he haa a high sig level. You will likely need either great skill with a Mystic or a champion with access to slow, for example She-Hulk or Stealth Spider-Man. That fight won't be easy by any means, but without an Unstoppable counter it's next to impossible.
6.4.6 Grandmaster: Hoo boy. Make sure you watch a guide or something to understand what his different phases are, but remember this: There are no amount of guides that can prepare you for going in and doing it yourself. This fight is definitely one that's easier watched than done. I recommend champions with high special damage to nuke him during stun (Shang-Chi, Kitty, Herc). Also, a reverse control immune champion is not strictly necessary, but it makes his third phase MUCH easier. A large part of the fight is not who you are using but your skill in doing his challenges. In my opinion, this is the hardest fight in the game in story mode. Good luck, and save some units before this fight just in case.
Then focus in the variants. They will greatly deepen your 5 and 6 star roster and give a lot of rank up materials. With that new deepened roster you will have a far easier time doing the rest of act 6 to get thronebreaker.
My suggestion is learn the counters. Act 6 cannot work without counters. See some below that I used. Your journey will be different.
1. Nimrod, Hela and Heimdal was used for Mr. Sinister boss in 6.22
2. Magneto basically ran through several bosses by himself
3. Hit Monkey was needed for 6.3.6 Cap IW to guarantee crits
4. Rintrah made easy work of 6.2.6 Champion. I one-shotted him and he was actually one of the easiest bosses
My suggestion is read guides on all act 6 bosses and make a list of counters. As you get those counters, rank them up and then when comfortable you should do the thronebreaker run. If it takes time its ok, nothing great offered in thronebreaker that you can immediately take advantage of. The Carina challenges and modes that come with Thronebreaker are anyways very high skill level, so rushing it will not benefit you.
