WHOM SHOULD I FOCUS TO RANK-UP?

I have 5* Hyp, 4* (Antman, Wasp, Colossus, Ultron, Dr. Octopus, Mordo), 3* (Blade, Domino, Gwenpool, Massacre, Phoenix, Drax, Cap America original, Ghost, Morningstar, Darkhawk, Taskmaster, Korg, Storm, Psylocke, Loki, Yondu, Green Goblin, Red Skull & some more). Now I am Level 40 & in Act4 Chapter 2 .

1. What should be my mastery?
2. Which Champions should I rank up or max out to get 2 Team &
3. Choose which champions so that I get some synergy or good combination in each team?

    Focus on your 5* and then the 4*s (Ultron, Doc Oc, Mordo, Wasp) leave the 3 starts alone unless you play area you'll never use them
