Who should I focus on upgrade

I’d mainly focus on 6*s until you can’t anymore. Shuri, captain marvel, guardian, and red skull are all very good and will cover a lot of ground for you.

If you have the time, I’d also upgrade that 5* hulk. He gives you a great science options which you currently don’t have right now.
