As I'm a newer player and only have 7 champions that are 4* and level 3 or 4. Of those some are better than others to play.

Gwen pool.
Ant man - level 2 just got him.
Which should I focus my better players on.
Defending or attacking....
Does it matter?
Which better benifits me attack or defense and which would benefit an alliance..

Most of the time, I am in top 3 (mvp) but I am unsure if there are really any tangible benefits.
At lower AW tier (which I assume your alliance are playing), here are some suggestions (and I think other forum member may have better insights).
Unstoppable Colossus
Can go on tile 11 [resistance]
Quake
Can go on 12 or 23 [outlast]
Old Man Logan
Can go on tile 2, 7, 20, 24, 34, 52 [fury] or 22 [true strike]
Hood
Can go on tile 13, 25 (13 also gives 2 bar power at start of fight) [special 2 bias]
Thor
Can go on tile 18, 33 or 42 [armor break]
Black Widow
Tile 24 [abilities]
Gwenpool
Can go on tile 6, 17 or 29 [bleed]
Ant-Man
Maybe tile 4 [poison] and if awakened , tile 40 or 45 [thorns]
Check it out if it works and these are not the only tiles champs can go onto. Also, at higher AW tier, there linked nodes giving more variations.
