It depends on you. A 5* takes more iso and more resources to rank up.

Yes. Her utility can be used against a wide range of champions.

ROL is doable with a 3 star gilly as well, mystic is the strongest class in the game save those t4cc for someone like Voodoo, Magik, SW or Ghost Rider. I have guy in my alliance who did ROL with Rouge at R4, she is pretty great plus awesome prestige.
