Nope, beating a different difficulty only re-rolls the boss for THAT difficulty. So, yes, you will have to beat Emma, and then hope re-roll results in different boss next time on that difficulty. But…1 -

FYI, there is NO DAILY RESET (if you can’t get past one). Just tested it and Boss and Hidden remained the same starting new quest again after the time when new Objective came out. So you just have to…3
