I'm sure our community could help weigh in with their opinions. As Namelez says, it could depend on the 4* Champions that you have at the moment, as focusing on a handful may help get you through tougher content quicker if you learn the way they work and how to use them most effectively.
This is a solid strategy, and can get you the rewards sooner for completing harder Quests, so that you can continue to expand your roster overall.
On the other hand, if you spend a bit of time on Arenas, you may find that working on your 3* Champions might be beneficial, as stronger Champions mean faster fights overall.
As a personal example, I spend quite a bit of time in the Summoner Trials arena for the Chips, and all of my 1* and 2* Champions are maxed out.
This means that while my reward potential is lower (Relative to a 4* Arena for example), I can get through fights incredibly quickly and get myself to meet Rank with less effort, owing to my Champs being tough enough to get the job done faster.
As Namelez said, it really does depend on your personal playing style and who you have as a 4* right now. If you're likely to spend more time in Arena than Quests, then it might be worth looking into working on your 3* Champions for the Arena Points.
With that being said, if you're looking to get through Quests with much tougher Champions to get those sweet rewards, then going straight for the heavy hitters and investing in your 4* Champs first might be a better option.
Start with x23...her regen helps clear so much content...I used mine(4* 5/50) in my runs to become uncollected..don't use her specials and let the power bar fill up..the more power she has,the more she will heal once her regen procs
