Colossus is up there surely. A r5 can definitely hit a 350k sp2 without suicides or boosts but with synergies. That would be at circa 50 armours and obvs you’re relying on every hit being critical.
If you’re playing well with the armours and can land one around 80 you could probably hit closer to 450k, the first is based on what I’ve actually done myself.
Put suicides on and that skyrockets.
A lot of the champs here at r5 can get nowhere near to those numbers.
As others have said, it's a very situationally based question. Factors like the champ it's being used against, and what you define as the special (just the immediate hit only, or the residual effects of that attack).
I'm choosing Longshot though, based on the potential of his sp2. It is obviously very dependent on using it against a buff heavy champ, but I have melted cosmic champs in Act 6 with a single sp2. Note, it's not all necessarily from the immediate damage, but if the defender continues to activate buffs while his fate seal is active, then they continue taking damage from what was effectively his sp2. Same with continuing to apply bad karma to a defender while the sp2 fate seal is active.
sunspot, capiw, proxima massive sp2..but all condition based....sunspot if he has 3-4 solar flares bye bye massive health pools...capiw if he has 5 charges and inflicted like 3-4 armour breaks and has that passive fury game over...proxima all challenges complete 100 plus combo ...boom...FATALITY!!!
Guardian. At 5/65 or Rank 2, he can do almost 3 million damage on an SP2 if every hit crits (see data here). This video shows him taking out ~1.5m X-23 in one go.
Obviously this very impractical and in any normal gameplay he will not do this amount of damage... but you asked for the biggest SP2 that is currently attainable in the game and this (or Morningstar with a crazy number of buffs) is probably the answer.
EDIT: The SP2 is at the beginning of the video so you don't have to watch all 10 minutes.

Because her damage is burst damage, both the sp2 as pictured and for some hits afterwards she was doing 20-30k hits maybe 5-10 seconds.
She’s pretty hard to ramp up, within that 200 hits I probably re-started 3 times.
You have to fight entirely via intercepting, no blocking, no parry. I blocked a couple hits along the way by mistake.
I would estimate I could do the fight as r2 in 100 hits maybe less, could work it out from the video.
Bear in mind this r2 is equivalent to a r4 4*.
The reality is that it’s not super practical but it’s a cool thing to pull off. Kinda like the guardian sp2 I guess.
There are synergies that act as a fail safe to protect losing the damage ramp up so I’ll take her vs ROL wolvie soon and see what she can do as r3 or r4.
