Should I upgrade or ascend any of these champs?

Where are you in the game? That makes a big difference.

Knull has got a lot of good utility, anti-evade mechanics, armour breaks, massive damage.
Long Shot is frankly amazing and can decimate opponents, particularly with his L2
Thing is more defensive but has some great utility with his immunities.
Spider-Man (Stark Enhanced) is an absolute nuke offensively if you can get the hang of building his poise charges and keeping them topped up.
Black Widow (Claire Voyant) has got amazing utility and is only available as a 6* so you have no concerns about being made obsolete by eventually pulling a 7*. She can switch immunities between Bleed, Poison, and Incinerate, reliably nullify buffs, and can also Regen, Power Drain, or deal burst damage respectively between those 3 stances.
Hit Monkey is also not available as a 7* and is fantastic - guaranteed crits and massive bleed damage. MLM and watch them bleed.
Dust I don't know much about offensively, but she's a great defender, same with Iron Man (Infinity War).
Peni Parker has got some amazing utility, especially if she's awakened and against someone with regen. Regardless, if you can build her charges and nail them with an SP2 they just burn to a crisp. Also a great defender.
Wong is more for long-form content but also a good defender.
Jabari Panther people are generally luke warm about, but she's also got good anti-evade utility and reliable cleanse of debuffs.
Wiccan I personally absolutely love - if they gain buffs, he can burn them to a crisp; it's honestly funny fighting the likes of Hulkling or Gorr with him because the fight's over in literally seconds.
Spider-Man 2099 is buff immune and has some great abilities
Proxima is also more for long-ish form fights and is kinda tricky with not amazing payoff.

So...depending on what you need, I'd recommend ranking up any of Knull, Long Shot, Spider-Man*, Black Widow, Hit Monkey, Peni Parker*, or Wiccan*. (those marked with an * are somewhat skill-based in using them right, meaning you can't just parry+attack to get results out of them.

Happy to help - good luck in progressing. It's well worth charging through the story content to get the better rewards, but don't bother trying to explore Act 6, just do a completion run and merrily skip to Act 7.

Take up ht for sure. I used my 5* all the way till act8. Then Claire is awesome. 3 immunities on demand, and powerful power control. She was my mvp in act6. Still hunting for 7* Claire. Hitmonkey is kinda glass cannon. U hit super hard but if u get hit, it's over. So u can do him too.

And as said by Panther, Longshot, knull, wiccan are also awesome. But I would recommend ht and Claire before them.
