Nick Fury is easily the best option there. His sig level doesn't matter - he just needs to be awakened.

My 6* r2 Wasp is ridiculously good. Used her to (occasionally) clear 50%-90% of a lane in act 6. She just becomes a damage monster once you learn how to heavy spam in the corner, and you don't have to worry about her low health.
