    All but tech get their attack reduced by 50%.
    So there are pretty much 2 options for you. Either use tech champs. Green Goblin and Stark Spidey in your case.
    Or use the champs you play best with, and just accept it will be very long fights. Doom is pretty good for longer fights because of the power control.
    Bring your corvus and take whatever charges you can to use him against howard.

    Every quest in 5.3 has a 50% reduction for all but 1 class. So it would be easiest to bring up 2 of each class to atleast 4* 5/50 or 5* rank 3 level. Otherwise just expect long fights.

    Honostly, when you are in act 5 it really still comes down to who you play best with. It doesn`t really matter.
    You could look up tier lists like KT1's on youtube. But I would really suggest to pick the champs you like.
    personally I would choose the following:

    skill: blade, shang chi
    cosmic: Corvus, Medusa (great for the final boss of act 5)
    science: Quake, she-hulk
    Mystic: Doom, Sym Supreme
    Tech: Stark Spidey, Ultron
    Mutant: Colossus, I would also rank Proff X, but since Iceman is already ranked up, he and colossus make a good duo aswell.

    again, that is personal preference.
