For R4 options! No where close to R4 any 6* yet but I would like to know who are the top 5 of each class that is worth of being R4!. When the time comes I'll know who to focus on

Mutant - Kitty Pryde
Skill - Nick Fury
Science - Human Torch or CAIW, but i might go with Miles for fun.
Mystic - Doom / BWCV, but i might go with Tigra for fun.
KT1 has a bunch of real nice tier lists, including an overall tier list and one for each respective class. He does a great job justifying his picks.
Skill: Nick, Kingpin, Shang Chi, Ragnagod (for Prestige), maybe Mole Man
Mutant: Kitty, Pr X, Toad, Sauron and Archangel
Tech: Ghost, Nimrod, Omega Sentinel, Warlock, Peni
Cosmic: Herc, Corvus, Knull, CGR, Annihilus (yes Annihilus)
Mystic: Doom, Tigra, Claire, Diablo, Swigil
Science: iBom, Torch, Piggy, Scorpion, Mr. Negative
Some of those that I threw in there will certainly trigger some disagrees lol
In my view, the best choices are all just your favourite champion.
At this point, everyone knows the good champs and who is great. But we don’t have hard enough content to need a rank 4, the only place you need one is in AW to reduce chance of death, and if you don’t take that seriously then the extra stats from R4 should only really get to go to a champ you really enjoy.
So for me, unless you have a specific champion you want to rank for AW and you use them all the time, just rank who you enjoy the most. Maybe you’re path 9 and want a Kingpin, or you’re path 7 and you want Spidey 2099.
But if you’re not taking AW that seriously. Rank Spidergwen if she’s your favourite champion, rank Sauron if you love dinosaurs and think his kit is fun. This game is to have fun, and ranking cool champs is fun.
Equally, if your favourite champion is Herc, rank him too! If you need prestige rank surfer. As long as they’re useful or you’ll use them or you want prestige, rank whoever you want!
With that said, here are my first rank ups of each class I’d do.
Skill: Killmonger
Science: Negative
Mystic: Tigra
Cosmic: Herc
Tech: Nimrod (or Omega sentinel if I pull and like her)
Mutant: Apoc (or Havok, I’ve just taken him to R3 and want to test him out)
Science: Immortal Abomination (already at r4).
Mystic: Tigra (already at r4).
Mutant: Apocalypse or Kitty.
Skill: Probably Kingpin, but Jabari Panther is an option in order to r4 the whole Jabari/Tigra/Kitty synergy trio.
Tech: Probably Nimrod, but Warlock and Captain America (Sam Wilson) might be good options in order to boost their mid-range damage.
Cosmic: I don't have any cosmic champions that I would like to r4 at the moment. Possibly Knull if I can awaken and max-sig him. A high-sig r4 King Groot would just be even more immortal than he already is.
Science: SM 2099 (already R4), MrN (if awakened), Torch, YJ
Mutant: Apoc or Kitty
Cosmic: Herc, Hypo (if awakened)
Tech: Nimrod, Guardian
Skill: Nick, KP, Aegon
Mystic: Tigra, Doom, Swigil
I’m not including champs I don’t have at all as 6* (Ultron, OS, Corvus, Diablo) who might be considerations. Deciding on #2 and will probably go with Tigra or Herc.
