It's referring to the node immediately before the boss where you can swap a champ out for another champ. For example, clearing the entire path with Hercules and then swapping him out for Kamala. Just bring in your Kamalas, don't swap anything, and you'll be good.

Ultimately, all you get for bringing both is one revive. You won’t be using team revives in this content because it’s only two champions, and you’ll end up reviving whichever one based on which is higher rank/sig/your favorite border color. If you’ve got both already ranked up, go for it. Otherwise, if you’re worried about it not counting, I’d just bring one.
