Cosmic and mystic are pretty obvious, but science has sentry, void, hulk and immortal hulk who are all stupid strong. I would honestly argue that immortal hulk is top 5 strongest in the game canonically

The question asked canonically in the comics and that has to be cosmic. You have all of your celestial gods, Odin, Galactus (here, Galan), Ego, Eternity, The Celestials, Zeus, The Living Tribunal, even Hercules and Thor (though, he and Odin are technically aliens viewed as the Norse gods, but that's probably changed by now), and powerful entities like The Phoenix (Force), etc. Then you have powerful beings such as Thanos, and Maestro, Silver Surfer, Gladiator, Adam Warlock, Ronan the Accuser, Annihilus, and many others. Symbiotes and any alien class falls under this class. I mean, it's the universe--it's huge! Of course it'll be the most powerful! Cosmic class can be multiversal as well. The next most powerful should be mystic. That class has powerful beings close to cosmic but not on the varied level. That class is also multiversal or multidimensional. Beings like Dormammu, Cleou, Doctor Strange, Wong, Doctor Doom, Mephisto, Nightmare, the Ghost Riders, and more are very powerful. Probably tied with mystic is the science class. The science class can be multiversal/dimensional as well. Heavy hitters like Sentry, Blue Marvel, almost every gamma related hero or villain, most spider-verse heroes

Spider verse aren’t at the level of the rest though
The only answer is mutant since it's the only actual class that's ever talked about. There are references to cosmic beings but mainly just suggested as from "other planets".
Canonically no class is the best, for two reasons.
First, there’s no such thing as class in the comics. Class is not something that exists that the game adapts, class is something the game makes up completely out of whole cloth. Yes, there is magic and technology and mutations in the Marvel comics, but these don’t work like classes do in MCOC. Such things are not exclusive in the comics as they are in the game. Many Marvel characters gain their abilities from what we’d call multiple class sources. Dr. Doom is magic, science, and technology in equal proportions. That’s what makes him so powerful and dangerous.
But the bigger problem is what we might call the singularity problem. The Marvel universe is a fantastical place where anything is possible., In MCOC there is at least some semblance of distinction between the classes. Skill means this, mutation means that. We expect Science to produce certain things, and Magic to produce different things. But in the Marvel universe, every source of power has no limits. The science fiction author Arthur C. Clarke once wrote “any sufficiently advanced technology is indistinguishable from magic.” In the Marvel universe, any sufficiently advanced anything is indistinguishable from magic. Push Science far enough and you get World War Hulk and the Void. Push mutation far enough and you get Legion and Franklin Richards. Even skill can be pushed to fantastical places, the Chaste (i.e. the enemy of the Hand) was able to lean to manipulate life energies in ways that could even bring back the dead in some cases.
At some point, every color on the class wheel gets bright enough that it basically looks white. Having said all of that, and acknowledging that there’s a lot of inconsistencies and ambiguities in the writing, I would say that The One Above All writes the rules of the multiverse with a Cosmic pen. It is the cosmic entities that govern and rule and possess most of the top tier power in the Marvel Multiverse. But Magic is what he uses when he wants to break his own rules, or rather write exceptions to them. The Ghost Rider is just the Spirit of Vengeance, period. You don’t ask how powerful he is. He’s the spirit of vengeance. That’s his role in the Marvel multiverse, and power levels has nothing to do with it. If he had judged World War Hulk guilty, he would have acquired whatever power was necessary to defeat him., He didn’t, so he didn’t, and he left.
As an aside, in the Marvel Universe as I understand it, you can argue there are four classes of power. There is power that comes from the way the “physics” of the universe works. That’s science and technology, and to some extent skill. Then there’s power that comes from your own life force, aka Chi. That’s where the supernatural powers of the martial artists like Iron Fist comes from. Then there’s power that comes from the structure of the universe itself. That’s the cosmic beings like Galactus and the heralds imbued with the Power Cosmic. And then there’s power that is bestowed from other beings. That’s where most of the traditional magic comes from (and yes, I know that in the current Marvel universe there’s more flavors of magic than Ben and Jerry’s, this is just broad strokes). It is an interesting question to ponder, if the only difference between Iron Fist and Galactus is that Galactus taps into an infinitely larger bucket of Chi tied to the cosmos.
