Last one I consider to be a new game mode is dungeons will there ever be another new original game mode?

Can't be done. They would've had to make the game differently from the outset. Besides which, the game as constructed only works because the AI is stupid lol
I really want a champion origin story type quest. Like, a quest is added where you can only play as said champion (let's say Captain America) and all the nodes are designed keeping that champion in mind.
You bring in that particular champion, the quest includes all the champions connected to the character and the final boss is the arch enemy of the champion (in this case it'd be Red Skull).
The interesting part here is that the entire origin story of a particular champion is explained in the quest through dialogue and interactions. I feel like the individuality of champions often gets sidelined in this game, this could be a good way to fix it.

The problem with something like that taken at face value is the huge difference in damage output across the playerbase. You realize that a single Abyss run is comparable in total damage output to a 50 million health boss. An alliance of players capable of doing an Abyss run could probably collectively knock out 100 million health in a couple hours. Meanwhile an alliance of more casual lower progression players might be incapable of generating that much damage on a single target in the playtime available to them in an entire month without sacrificing doing normal game activities.
So if you do it globally the top 1% of 1% of players are going to disproportionately snag all the rewards. If you do it by alliance then most alliances will simply fail or the top alliances will auto-fight through it, depending on the size of the challenge. And if you scale the challenge by alliance rating or prestige, that's going to encourage all sorts of manipulation.
Training Mode, or Danger Room—whatever you want to call it. It would be nice to be able to test interactions between abilities and something like a “node select” function to allow the player to simulate fight conditions.
Also would help players understand how reliable their own champs’ abilities are in certain situations (e.g., does KP/CB/AV shrug off work like it’s stated for some debuffs but not others? or how does VtD toxic armor work against a champ like Korg?).
