So I thought I could try road to the labyrinth chapter three round one I have a little less prestige but higher level and rank with higher damage and slightly less hit points. And I got slaughtered badly. In almost every fight regardless of class advantage. The other chapters were a little easier. Is there some type of ai that I have yet to experience or is there something about these chapters I'm missing.

Screenshot those buffs on UC. That will help you know what to counter him with.
Chapter 3 has Safeguard. You need a Champ that can bypass it. BW, CB, Elektra, Falcon.
And to answer the OP's question, it's not an AI, its the node Safeguard on every champion, preventing each enemy from losing more than 1% health from a hit.
Safeguard is considered a defensive ability: it is like a special blocking power that reduces incoming damage on each hit. As a result, anything that debuffs defensive ability accuracy can reduce the chance of safeguard triggering from 100% to something less than that. If safeguard doesn't trigger, your damage lands at full strength (minus other defensive powers of course).
See the "5" on the right side? That number represents how many buffs the node that Colossus is standing on is giving him. Tapping should show you what those buffs are. It should include Safeguard which states "Prevents champion from losing more than 1% Health from a single source". "Single source" generally means "in one hit."
Scouting the buffs that are active on a node is an important element of the game to learn, because it will become increasingly important in higher end content. I'm kind of surprised this is something you haven't come across yet. Beyond a certain point, most of the difficulty in the content will come as much from the node buffs as from the specifics of the champion you're fighting.
Champions such as BW (duped), Elektra, Crossbones can bypass the Safeguard or you could go the route of damage over time by using bleed champs (Black Panther, Wolverine)
This chapter can be completed with 3* 4/40 champs as well with little ease. Wish you the best of luck!
Ability accuracy debuffing champions are not necessary to complete RTTL 3.1. They would make it easier for you to complete it. Honestly, I think you are attempting content far above both your roster strength and your current skill level. You might want to consider completing Act 4 and doing Realm of Legends first. While you are currently in the relatively easy part of RTTL, you are attempting to complete the third (or so) hardest content in the game (behind Labyrinth of Legends and Act 5.2).
Also I've been playing for almost two years and I'm currently on RTTL 4.5.
